Research on Cabbage Simulation in Artificial Climate Incubator

High temperature is a common stress factor in summer and autumn vegetable production, which seriously affects the yield and quality of vegetables. The breeding of heat-tolerant vegetable varieties has always been one of the important tasks of vegetable science and technology workers in China, and important progress has been made. Some methods for identifying heat resistance of different vegetable crops have been explored. At present, the early autumn cabbage varieties planted locally in Wuhan are dominated by strong 50 imported from Japan. The varieties have strong heat resistance, early ripeness, green balls and good quality, but their ability to resist cracking balls is poor. Through nearly 10 years of work, the Wuhan Vegetable Institute has selected several new varieties of early autumn cabbage with independent intellectual property rights and good comprehensive traits that can gradually replace imported seeds. To test the resilience (heat resistance) of these new varieties, this experiment was carried out on the basis of direct field identification. The artificial climate incubator simulates climate during the study of cabbage and facilitates research.

Select the robust leaves from which all varieties of cabbage are fully developed, wash them with deionized water, avoid the main veins, and use a punch to take 5 leaf disks with a diameter of 1 cm into a test tube, add 5 mL of deionized water, shake well, and then measure the initial Conductivity EC0, and then placed in a 48 °C constant temperature water bath for 30min, quickly cooled after taking out, left at room temperature for 2h, fully shaken, conductivity was measured with a conductivity meter EC1, and then the tube was placed in boiling water and heated for 20min, After taking out the rapidly cooled EC2, the final conductivity was measured and repeated 3 times.

The identification of the high-temperature form of sunlight greenhouse in summer can basically reflect the difference in heat resistance of cabbage, but it is difficult to distinguish the difference between heat-resistant and more heat-resistant varieties, and the high-temperature stress identification method of artificial climate box using yellowish wilt as a heat-damage index of cabbage leaves can be clearly reflected. The heat resistance of cabbage material was different, and it was determined that the treatment of cabbage seedlings at a constant temperature of 36.5°C for 6 days was a suitable pressure for selecting the heat resistance of cabbage.

The determination of artificial climate incubators found that, from the viewpoint of the heat resistance identification method of cabbage, the proline method is more complicated, the heat damage index morphology method is simple and intuitive, and the proline content determination method is combined with the seedling climate chamber stress method. Quantitative determination of the heat resistance of cabbage material can be made. The combination of the two methods can fully reveal the heat-resisting characteristics and heat-resistance mechanism of cabbage varieties and guide the breeding better.
